Advanced Training in Techniques of Epigenetic Regulation of Gene Expression
The proposed training program aims to enhance research skills in the field of epigenetics and gene
editing under the DBT-BUILDER program. The program includes hands-on training sessions that will
guide participants on performing advanced techniques, such as Chromatin-immunoprecipitation (ChIP)
assay, followed by quantitative real-time PCR (qPCR) to study specific gene promoter level
enrichment of transcription factors and histone modifications. The three-day training session covers
both the theoretical and practical aspects of the proposed assay, as well as the analysis of the
obtained data. On the first day, participants will learn the fundamental principles and application
of the assay system through research lectures by external and internal experts. Furthermore, on the
same day, the participants will start the hands-on training by performing the key part of the ChIP
assay, which will continue on the second day of the workshop. On the third day, ChIP-isolated
genomic DNA will be used to perform qPCR analysis using unique gene promoter-specific primers. After
completing the experiment, participants will also learn about the analysis of the acquired data. The
main objective of the proposed workshop is to train young scientists in advanced techniques like
ChIP assay and qPCR analysis, which will enable them to evaluate the promoter-level enrichment
status of transcription factors, DNA binding proteins, and modified histones, indicating their role
in regulating complex gene expression networks in both physiological and pathological contexts.
